چکیده

The aim of this study was to investigate the evolution of protozoa numbers in two types of rumensimulating fermenters [single-flow continuous culture fermenters (SFCCF) and semi-continuous flow fermenters (Rusitec)] receiving high-forage diets. Ruminal inoculum from sheep and goat were used in SFCCF trials, whereas only sheep inoculum was used for Rusitec fermenters. In both systems incubation trials were conducted for 14 days by following the methodologies usually utilized in each of the participant laboratories. The flow through all fermenters was maintained by continuous infusion of artificial saliva at a dilution rate of 5.3%/h (1.3 per day). Retention time of solid digesta was 19 and 48 h in SFCCF and Rusitec fermenters, respectively. Protozoa numbers decreased (P < 0.001) rapidly with time from the first day after inoculation in all fermenters. The decrease was more pronounced in SFCCF than in Rusitec over the first 2 days post-inoculation, with values dropping to 17.8 and 33.0% of those on day 0 in SFCCF and Rusitec, respectively. In contrast, on day 4 SFCCF maintained greater protozoa numbers than Rusitec fermenters (8.75 × 10 and 4.1 × 10 per ml, respectively). Protozoa completely disappeared after 7 days in SFCCF, whereas they were present in Rusitec fermenters even 14 days after inoculation (1.0 × 10 per ml). Holotrich protozoa disappeared completely in SFCCF by the second day after inoculation, but they were maintained in Rusitec fermenters for 12 days. The better ability of Rusitec fermenters than that of SFCCF to maintain protozoa might be related to the greater retention time of solid digesta in the Rusitec system.

Comparison of microbial fermentation of high- and low-forage diets in Rusitec, single-flow continuous-culture fermenters and sheep rumen.

Eight Rusitec and eight single-flow continuous-culture fermenters (SFCCF) were used to compare the ruminal fermentation of two diets composed of alfalfa hay and concentrate in proportions of 80 : 20 (F80) and 20 : 80 (F20). Results were validated with those obtained previously in sheep fed the same diets. Rusitec fermenters were fed once daily and SFCCF twice, but liquid dilution rates were sim...

Effects of defaunation on fermentation characteristics and biotin balance in an artificial rumen-simulation system (RUSITEC) receiving diets with different amounts and types of cereal.

Biotin is required by rumen microbes for efficient fermentation. To evaluate the role of protozoa in ruminal biotin metabolism, five diets composed of grass hay or of grass hay/cereal grain mixtures were supplied to faunated or defaunated RUSITEC fermenters. In the mixed diets, hay was replaced to 33:67 or 67:33 w/w on an air-dried basis by either wheat or maize grain in order to simulate diffe...

Utilisation of phytate phosphorus by rumen bacteria in a semi-continuous culture system (Rusitec) in lactating goats fed on different forage to concentrate ratios.

Experimental data on phytate phosphorus utilisation by ruminants are scarce. The aim of this study was to estimate the phytase activity of rumen micro-organisms when phytate phosphorus supply is high. A semi-continuous culture system fermentor (RUSITEC) was used. The inoculum was obtained from eight goats fed on either high or low forage level diets. Experimental buffers only differed by the na...

Effects of Thyme Essential Oil and Disodium Fumarate on Ruminal Fermentation Characteristics, Microbial Population and Nutrient Flow in a Dual Flow Continuous Culture System

The aim of the present study was to investigate the effects of di-sodium fumarate (DSF) and thyme essential oil (TEO) solely and simultaneously on ruminal fermentation properties and microbial abundance. A dual-flow continuous culture system (DFCC) with eight 1400-mL fermenters was used in a period of 12 d that divided to 9 d for adaptation and 3 d for sampling. Fermenters were fed 100 g d...

The Effect of Forage Level and Oil Supplement on Butyrivibrio fibrisolvens and Anaerovibrio lipolytica in Continuous Culture Fermenters

The objective of this study was to evaluate the effects of forage level and oil supplement on selected strains of rumen bacteria believed to be involved in biohydrogenation (BH). A continuous culture system consisting of four fermenters was used in a 4×4 Latin square design with a factorial arrangement of treatments, with four 10 d consecutive periods. Treatment diets were: i) high forage diet ...
